Is it possible to setup git annex to make a repository out of an already existing folder with files on Google Drive?

Not yet. What you're looking for is import tree. It's currently only available for Amazon S3 and directory special remotes, afaik. As soon as it's added to the external special remote protocol, I'm going to implement it.
