-I’m currently a Ph.D. student in Electrical Engineering at Yonsei University advised by [Prof. Seung Ah Lee](https://sites.google.com/oisl.me/oisl/) with my research centered on computational optics and imaging. During my Ph.D. studies, I had the opportunity to be mentored by [Prof. Roarke Horstmeyer](http://horstmeyer.pratt.duke.edu/) in the Biomedical Engineering department at Duke University. I've been honored to receive the Silver Prize at the [28th Samsung Humantech Paper Awards](https://humantech.samsung.com/saitext/index.jsp) and Graduate Research Fellowships from the [NRF](https://www.nrf.re.kr/eng/index) and [MOHW](https://www.mohw.go.kr/eng/index.jsp).

-We report tensorial tomographic Fourier ptychography, a nonscanning label-free tomographic microscopy method for simultaneous imaging of quantitative phase and anisotropic specimen information in 3D. Built upon Fourier ptychography, a quantitative phase imaging technique, T2oFuT2oFu additionally highlights the vectorial nature of light. The imaging setup consists of a standard microscope equipped with an LED matrix, a polarization generator, and a polarization-sensitive camera. Permittivity tensors of anisotropic samples are computationally recovered from polarized intensity measurements across three dimensions. We demonstrate our system's efficiency through volumetric reconstructions of refractive index, birefringence, and orientation for various validation samples, as well as tissue samples from muscle fibers and diseased heart tissue. Our reconstructions of healthy muscle fibers reveal their 3D fine-filament structures with consistent orientations. Additionally, we demonstrate reconstructions of a heart tissue sample that carries important polarization information for detecting cardiac amyloidosis.

Smartphones and mobile devices can be useful for the construction of compact and portable imaging devices, by replacing expensive and bulky optical instruments with state-of-the-art cameras, high-performance processors, and network connectivity in a compact footprint. The rapid dissemination of smartphones and their user-friendly interfaces can allow for microscopic imaging and evaluation of biological specimens to be accessible to the general public as well as trained health care professionals. As the technologies for image sensors, miniaturized optics, and mobile processors continue to develop, these smartphone-enabled devices are expected to change how medical tests and evaluations are conducted in the field, as well as in the lab.

-FPM reconstruction process heavily depends on the fast Fourier transform for propagating the complex field back and forth between the sample and the image plane. The algorithm is implemented on our custom Android application using the open CV library. In the application, we recover about 150 µm x 150 µm region at a time, which takes about 30 seconds including the loading time of the saved images.

-We applied our system to observe a biological sample. We imaged unstained blood smear with our device and acquired a high-resolution image over a large area as shown in this slide. For color FPM imaging, each color channel is captured and reconstructed separately. The full FOV reconstruction result of the Pap smear sample shows a much sharper features of the sample.

-Advances in image sensor technology and miniaturized optics have enabled the development of ultra-compact and high-quality cameras for everyday use. However, while the pixel resolution of cameras continues to increase, there are physical limits to reducing the size and cost of the entire camera module. As an alternative approach, various lensless imaging methods, which replace the lenses with thin light-modulating masks, have been recently developed. In lensless cameras, the entire module can be miniaturized by reducing the thickness of the lens elements as well as the focusing distance of the lenses. -In these lensless cameras, the overall field of view (FOV) and image quality are primarily determined by the light modulating properties of the phase mask originating from the locally varying microscopic structures on the mask. The mask and the resulting PSF inherently define the transfer function of the imaging system; further, the contrast and density of the PSF affect the multiplexing capability and reconstruction quality of the image under a finite bit depth of the image sensor. Inevitably, the ability to design and generate phase masks and the resulting PSFs are crucial for manufacturing and deploying lensless cameras for various applications.

- Fourier ptychography (FP) is a powerful computational imaging technique that provides both super-resolution and quantitative phase imaging capabilities by scanning samples in Fourier space with angle-varying illuminations. However, the image reconstruction in FP is inherently ill-posed, particularly when the measurements are noisy and have insufficient data redundancy in the Fourier space. To improve FP reconstruction in high-throughput imaging scenarios, we propose a regularized FP reconstruction algorithm utilizing anisotropic total variation (TV) and Tikhonov regularizations for the object and the pupil functions, respectively. To solve this regularized FP problem, we formulate a reconstruction algorithm using alternating direction method of multipliers and show that our approach successfully recovers high-quality images with sparsely sampled and/or noisy measurements. The results are quantitatively and qualitatively compared against various FP reconstruction algorithms to analyze the effect of regularization under harsh imaging conditions. In particular, we demonstrate the effectiveness of our method on the real experimental FP microscopy images, where the TV regularizer effectively suppresses the measurement noise while maintaining the edge information in the biological specimen and helps retrieve the correct amplitude and phase images even under insufficient sampling.
